Abstract: The aim of this study was to standardize the 3 techniques designated as exotoxin ELISA, sonicated ELISA and dot-blot ELISA in the detection of antibodies to Corynebacterium pseudotuberculosis in sheep sera and to evaluate the usefulness of these techniques in the routine diagnosis and screening of C. pseudotuberculosis infection. A total of 471 sheep sera, 100 of which were from vaccinated animals, obtained from agricultural enterprises under the General Directorate of Agricultural Enterprises were examined. Among the 471 serum samples, 172 (36.5%), 119 (25.2%) and 181 (38.5%) sera were positive by the exotoxin ELISA, sonicated ELISA and dot-blot ELISA techniques, respectively. The results obtained in this research showed that exotoxin ELISA, sonicated ELISA and dot-blot ELISA techniques are useful screening tests for the routine diagnosis of caseous lymphadenitis disease in sheep.
