A New Method for the Purification of Carbonic Anhydrase Isozymes by Affinity Chromatography

Abstract: A new affinity chromatography for purification of carbonic anhydrase isozymes was prepared by using Sepharose 4B as a matrix, L-tyrosine as a spacer-arm and p-aminobenzenesulfonamide as a ligand. The binding capacities of the affinity gel were determined at different temperatures, pH's and ionic strengths. As a result of the affinity gel having good flow properties and spacer arm, human carbonic anhydrase isozymes (HCA I and HCA ll) and bovine carbonic anhydrase (BCA) were purified in high yields from erythroyctes.
