In-Vitro Regeneration of Sesame (Sesamum indicum L.)

Abstract: Effects of various plant growth regulators on organogenesis from cotyledon explants of Sesamum indicum L. Muganli-57 were investigated. Murashige and Skoog medium supplemented with 1, 2, 4, 8 mg/l BAP and 0.1 mg/l NAA or without NAA was tested. Callus formation started in ten days and a high frequency of callus formation (up to 100%) was obtained. Adventitious shoots were obtained in an MS medium containing 4 mg/l BAP and 0.1 mg/l NAA or 8 mg/l BAP and 0.1 mg/l NAA after 65 days in culture. Organogenesis occurred at the cut end of the cotyledonary explants. This regeneration system can be used for Agrobacterium-mediated transformation of sesame.
