Purification and Characterization of Dehydroascorbate Reductase from Parsley Leaves

Abstract: Dehydroascorbate reductase, the enzyme that regenerates ascorbic acid in several tissues, has been purified from parsley leaves. The purification steps included Ammonium sulphate fractionation, Sephadex G-100, CM-Sephadex chromatographies and Sepharose-GSH and Hexyl-GSH Sepharose affinity chromatographies. At the end of the purification steps the enzyme had been 976 fold purified. The molecular weight of the enzyme is 24.000 dalton. It has been found that one molecule of the enzyme contains six sulfhydryl groups. Also, some biochemical properties of the enzyme have been determined.
