Abstract: The aim this study was to determine the capability of embryos from an outbred OF1 mouse strain to form ES cell-embryo chimeras with morulae injection. When the R1 ES cell line was routinely cultured on mitotically inactivated feeder cells in a culture medium supplemented with Leukemia Inhibitory Factor (LIF), the colonies maintained undifferentiated ideal ES cell morphology. When ES cells were injected into 55 eight-cell embryos, 73% of the embryos combined with ES cells. 31% of the embryos transferred to recipients developed to term. 27% of offspring were chimeric. ES cells contributed about 90% of the coat in the chimeras. This study demonstrates. that high frequencies of chimerism can be obtained by injection of ES cells to morula from the OF1 outbred strain.
