Comparative Replication of Autographa californica Nuclear Polyhedrosis Virus in Insect Cell Cultures

Abstract: In this study. using Autographa californica nuclear polyhedrosis virus (AcNPV)-5podoptera frugiperda (Sf) infection as control. the comparative replication of AcNPV in Anticarsia gemmatalis (Ag) cells was investigated by Iight microscopy. SDS-polyacrilamide gel electrophoresis (PAGE) and cell culture infective dose 50 methods (TCID{_50}). As a result of these studies. it was observed that the cytophatic effects in AcNPV-Ag infection appeared about 12 hours Iater as compared that of Sf cells and these effects occurred in about 80% of cell population. The proteins which are specific for virus were produced at about 50% Iess and 12 hours Iater. The amount of progeny was observed to be dramatically Iow (90%). and the production period was delayed 12 hours. too. The evidence indicating host range and expression feature of AcNPV, has shown that the use of Ag cells in expression studies is not very efficient. however . this cellline is permissive for AcNPV replication.
