An Investigation on In Vitro Regeneration Ability of Madder (Rubia tinctorum L.)

Abstract: In this thesis regeneration ability Rubia tinctorum was investigated by using tissue culture techniques. For this aim. the explants from node. internode, leaf and hypocotil were cultured in Murashige and Skoog (MS) medium, containing different concentrations of NAA and BAP. Anthers were placed in NN medium containing different concentrations and combinations of NAA. BAP and different concentrations of sucrose. No regeneration was seen in anthers. Node and hypocotile produced shoots from meristem and buds. On the other hand. internode and leaf explants produced callus which later produced only roots. This study showed that regeneration ability of the oversoil parts of madder plants could be used practically in tissue culture. In callus and roots- colouring varied from yellow to brownish red in terms of anthraquinone synthesis.
